The detailed operation process of the fermentation tank is here


Fermentation tank refers to the device used for microbial fermentation in industry. Its main body is generally a main cylinder made of stainless steel plate, with a volume ranging from 1m³ to several hundred m³.

 

In design and processing, attention should be paid to the tight and reasonable structure. It should be able to withstand steam sterilization, have a certain degree of operational flexibility, minimize internal accessories (avoid dead corners), have strong material and energy transmission performance, and be able to make certain adjustments to facilitate cleaning and reduce pollution, so that it is suitable for the production of various products and reduce energy consumption.

 

1. Preparation of empty tanks: Remove the fermentation tank, clean the fermentation tank, load the material, cover the tank cover tightly, and tighten the bolts on the tank cover. Calibrate the pH and DO electrodes, insert the electrodes into the sensor interface on the tank cover and tighten the nuts in time. The stirring motor interface, pH, DO, and foam electrode interface are all wrapped with tin foil; install the air filter membrane at the air inlet, and clamp the connecting pipe with a clip; use a spiral clamp to control the appropriate opening of the tail pipe, and wrap it with eight layers of gauze and newspaper; close the sampling tube.

 

2. Sterilization: Place the fermentation tank in the autoclave for sterilization.

 

3. Cooling down: Put the sterilized fermentation tank back to the fermentation position, connect the motor and the electrode wires of pH, DO and foam electrodes, and install the temperature electrode into the temperature measurement interface. Connect the circulating water pipeline, turn on the circulating water switch, and set the temperature to automatic.

 

Connect the vent to the air filter, turn on the air flow meter, open the connection between the air filter and the tank, and let air enter the tank. Note: During the cooling process, always monitor the tank pressure and it is strictly forbidden to drop to zero.

 

4. Fermentation When the temperature drops to the required temperature for fermentation, it enters the fermentation stage

 

1) Set each fermentation parameter to the fermentation process value, and calibrate the dissolved oxygen electrode to 100%.

 

2) Inoculation: Lower the tank pressure to no more than 0.02Mpa. Loosen the inoculation port in advance, place a flame ring at the inoculation port and light it (add cotton to the ring to help combustion), open the inoculation port, pour in the seeds, then tighten the inoculation port and immediately increase the tank pressure.

 

3) Fermentation initialization: Enter the "fermentation batch number" in the "initialization" module and change the fermentation status to "start".

 

5. End of fermentation

 

1) When fermentation is finished, change the fermentation status to "End" in the "Initialization" module. Open the sampling valve and the tank bottom valve to discharge the material.

 

2) After discharging, remove the feeding needles and cover the plugs. Use distilled water to clean the tank, then add appropriate amount of distilled water to soak the electrodes (the electrodes can be removed and properly stored if not used for a long time). Close all open valves, return the controller to the main menu, press Ctrl+F6 to close the control program, and finally turn off the main power.
